Disclosing the Interaction Between Scml2 and Yap1 in Prostate Cancer

In this study, sex comb on midleg-like-2 (SCML2) was identified as a binding partner of the YAP1 protein complexes isolated from nuclei of prostate cancer cell lines. Both SCML2 and YAP1 are known to regulate cellular biology. Western blot analysis showed that AR-positive cell lines express elevated levels of SCML2, suggesting a possible link between androgen hormonal signaling and SCML2. Immunofluorescence imaging revealed that androgen hormone signaling promoted the subcellular localization of SCML2 and YAP1 compared with mock control. Proximity ligation assay demonstrated that androgen regulated the physical interaction between SCML2 and YAP1 proteins. GST-pulldown revealed that SCML2 and YAP proteins physically interact in the test tube. Cell growth assays suggest that SCML2 could contribute to androgen independence. These observations indicate that the interaction between SCML2 and YAP1 has a crucial role in prostate cancer progression.
